Potent gene delivery from fluorinated poly(ß-amino ester) in adhesive and suspension difficult-to-transfect cells for apoptosis and ferroptosis.

Tremendous efforts have been made to improve polymeric property in gene delivery performances, especially when obstacle of transferring gene construct into difficult-to-transfect cells occurs. Innovations in the area of fluorination and fluorinated compounds with biomedical potential in medicinal chemistry are believed to assist in the development of new therapeutics. Fluorine modified polymers have shown to navigate the gene transfection cellular barriers and promoted the transfection outcomes. Gene transfer into some liver cancer cells and human leukemia cells has always been a challenge. Here, by facile incorporation of a fluorine containing amine monomer, 1H,1H-undecafluorohexylamine, fluorinated poly(ß-amino ester) (FPAE) was synthesized to significantly improve the transfection performance, achieving high transfection efficiency of 87% and 55% in two representative difficult-to-transfect cells, HepG2 and Molt-4, which were cultured in adhesive and suspension condition, respectively. However, the potency of Lipofectamine 3000 was very limited. More importantly, functional studies revealed that FPAE can dramatically outperform Lipofectamine 3000 in delivering Bcl-xL and PKCßII to either provide the protection against apoptosis or promote the ferroptosis in HepG2 cells. This work facilitates gene therapies by overcoming biological barriers for targeting difficult-to-transfect cells and disease models when medically necessary.

Tear trough ligament reset as a new method for tear trough deformity and dark circles correction.

Tear trough deformities and dark circles are important parts of periorbital aging. In this study, the tear trough ligament was reset to correct the tear trough deformity, and its effect on dark circles was analysed. The medical records of 60 patients who underwent eye bag plastic surgery between January 2021 and February 2022 were analysed (30 had traditional eye bag plastic surgery and 30 had the tear trough ligament reset). The tear trough deformity score and the dark circle score showed that the corrective effect on the tear trough deformity in the reset group was better than in the traditional surgery group at six-month follow up (mean (SD) tear trough rating scale (TTRS) score = 4.82 (0.95) vs 5.92 (1.00), p < 0.0001; L* value 55.27 (2.90) vs 47.51 (3.00), p < 0.0001). The satisfaction of patients in the reset group was significantly higher than it was in the traditional surgery group (25/30 vs 18/30, p = 0.045). Tear trough ligament reset is a safe and effective surgical method of periorbital rejuvenation, especially for beauty seekers with obvious tear trough deformities.

Sub-brow lift combined with orbicularis oculi folding for periorbital rejuvenation.

The study aimed to evaluate a new method of periorbital rejuvenation in which periorbital muscle manipulation is utilised. Patients (n = 82) who underwent eyebrow lifting surgery in Yichang Central People's Hospital were retrospectively included. All the patients had drooping eyebrows and lateral hooding of the upper eyelids combined with a flat or depressed outer-upper side of the eyebrow arch. Data were collected on the position of the eyebrow relative to the eye before and after surgery and the postoperative satisfaction of patients. The study included 43 patients in the traditional group and 39 patients in the combined group. The postoperative results showed that the combined approach group had more aesthetically pleasing mean (SD) ratio of the position of eyebrows relative to eyes (1.6 ± 0.2 vs 1.7 ± 0.2, p = 0.021). Furthermore, the satisfaction of the patients in the combined group was significantly higher than that of the traditional group (37/39 vs 34/43, p = 0.036). The periorbital rejuvenation through the sub-brow incision, combined with supraorbital orbicularis muscle folding had more aesthetically pleasing ratio of the position of eyebrows relative to eyes and the satisfaction of the patients in the combined group was significantly higher.
